I think you would take it as the time to complete your first playthrough, but in a moderately focussed manner.

I was curious on this one because for a first run time that is crazy fast Gombol. From what I've seen of the xbox version the puzzles are all the same and the fact that it is on a xbox controller rather than a kb and mouse even my console mates agree slows you down a bit as it is trickier to pull off some of the trick shots.
I've just run through the whole game and taking into account I've completed all the least portal challenges and the majority of the least time challenges. I've just completed the game in 39 minutes. 26 minutes for the first 19 bits and 13 for the last bit. That is including the waffle time of Glados to chat at you at the end. I doubt this is the fastest I've done it as not played it in a while so am rusty and made a few mistakes here and there.
If you say you did it in 40mins then cool I bow to your portal might, just tres impressive. Unless I'm missing something and there is something that makes the xbox version shorter.

It's just easier to control on the xbox version I think, bought portal for the pc (since I wanted tf2, and the orange box was cheaper then buying them seperatly at the time) and I can honestly say, the PC controls are more akward then the 360 controls (might be because I was a massive console gamer at the time, playing only wow on the pc, then.)
